ANTIBODY SHARED RESOURCE

CORE LEADER:  Barbara Christy, Ph.D.  (christy@uthscsa.edu; 567-7227)
CORE PERSONNEL:  Diane Jones & Sherry Dodds (doddss@uthscsa.edu)

Services provided:

  1. Rabbit polyclonal antibody production.  The antigen for injection (such as a purified fusion protein expressed in bacteria)  is generally supplied by the investigator.  The cost for this service is $400 for 1 antigen (2 rabbits injected).  Approximately 4 mg. of the protein antigen is required before the rabbits are ordered.
  2. Mouse polyclonal antibody production.  The antigen for injection is generally supplied by the investigator.  The cost for this service is $200 for 1 antigen (3 mice injected).  These mice can be used later for production of monoclonal antibodies, if desired.  Approximately 1.2 mg. of the protein antigen is required before the mice are ordered.
  3. Mouse monoclonal antibody production. (Requires approximately 1.2 mg of protein in addition to the protein injected into mice for polyclonal serum above).
    1. Production of hybridoma lines from responsive mice
    2. Identification of potential positive hybridoma lines by ELISA,  Western blotting against antigen
    3. Single-cell cloning of a limited number of positive hybridoma lines

(Cost for entire process is $2,000)

  4. Hybridoma supernatant production and collection (price depends on quantity).
  5. Purification of antibody from hybridoma supernatant (price depends on quantity).
  6. Antibody isotyping. ($20 per antibody)
  7. Adaptation of hybridoma lines to serum-free or low-serum culture conditions. ($100 per hybridoma line adapted)
  8. Cryopreservation of hybridoma lines.  ($20 per 3 vials of one line)
  9. Cryostorage of hybridoma lines.  There is currently no charge for cryostorage of hybridoma lines generated by the core, as long as space is available.
  10. Affinity Purification of Rabbit Polyclonal Antibody now available.  $50 for column set-up (coupling of antigen to resin); $15/ml of antiserum purified.

In some instances, the core can provide advice or assistance to investigators in the design, production or purification of bacterial fusion proteins.  Other antibody-related services may be available upon request, as time permits.

**Prices quoted are for members of the San Antonio Cancer Institute.  Prices for non-SACI UTHSCSA faculty will be higher.
